I am building a sealed box out of MDF for my truck.  I will use screws and glue for the box construction will this be enough to seal the box itself or, do I need to put something around the inside to seal it?  The box will be a wedge shape with three seperate compartments.

Then do I need to use anything to seal around the subs themselves or is the flat surface good enough (I wouldn't think it would be)?

For anybody wondering I am using three JL 10W3v2's and a 500/1 amp.

Be liberal with the glue and make sure edges are all very tight when the glue is drying. I use a bunch of clamps to make sure that all edges are very tight. Glue and screws should be sufficient. If you want to make sure there are no leaks, then use silicone caulk around all edges. Another option is to spray the entire inside of the speaker with something like this: Damping Spray I've done this with a couple enclosures in a row now and I really like it. Quick and easy.
This speaker gasketing tape can be used to ensure an airtight seal around the subs themselves.

As far as sealing the box itself, i prefer Liquid Nails. This can be had at home depot for under $10.

Sealing the sub...a thin piece of foam would be good to use, some woofers either come with a pre cut piece of foam or a piece already stuck to the basket, make sure it isn't to thick otherwise you may warp the basket. once again kfr01beats me to it.

What is the best way to get the sealant on the front inside of the box just smear it around with your finger.

Also what if any do you reccommend for dampening material in the box?

I assume that my local JL dealer (They are a very high end custom shop) would have the dampening spary and speaker gasketing tape?

I wound up using my fingers, they work great, due to their shape.

They ought to give you the gasket tape, it is super cheap and you probably spent some money on that gear, so don't be shy asking for it.

When I built my last box I used ruberized spray under coating on the inside of the box. It worked rather well. I also used a professional carpenders glue on the panels. That box is air tight.
