Mono amp: 400W x 1 @ 2 Ohm
Sub: 12" dual 4 Ohm 400W

I know I need to wire as follows: posted_image

My question is:

Does it matter which speaker outputs on my amp I use? Obviously, there are 2 sets (L+ L- and R+ R-). Does it matter if I use only the left + and -, or should I run left + and right -? Is there a right or wrong way, or am I going to the same output (400W) either way?

If its a mono amp than it should have speaker terminals for speaker 1 and speaker 2 not left and right. But regardless as to how its labeled if its a MONO BLOCK amp then only use 1 set of terminals, if you bridge this amp it will damage the amp.

A mono amp only has one output. Mono = one. If your amp has multiple connection points, they are in parallel to the one amplifier output. So no, it doesn't matter. Just realize you cannot consider them as seperate outputs and if you connect another speaker to the amp, it will be in parallel with the first one so plan accordingly.

Thanks to you both. I'm only planning on running this amp to the one sub. Oddly enough, my amp doesn't have left/right or speaker 1, speaker 2, it's labeled as such:

- L + R and - L + R

Since it's a mono amp, why even have two sets of outputs? Could I also run one set of outputs to the one side of the dual 4 Ohm connector on the sub, and the other set of outputs to the other connectors on the sub? Is this just a waste of wiring since I can just connect the 2 +s on the sub with a much shorter wire?

Like I said, a mono amp has only one output. If it had both left and right outputs, that would mean it was a stereo amp. The reason there are two sets of connectors is simply for flexibility. Yes, you can connect one VC to one set of connectors and the other to the other if you want to. It is much easier to simply run one + and - wire from either set of connectors to your speaker, and wire the VCs at the speaker as shown in your diagram above.
