What Frequency to use when setting gains?

Does it matter what frequency I use to set the gain of my subwoofer amp. I know it has to be below 80 Hz so the LP filter doesn't kick in, but does it matter if I use a 50hz tone, 70 hz, 30 hz, or what frequency of a tone. Does it matter.

50 Hz will be about as robust as they get.

I feel like an idiot. My mind blanked I had to look robust up posted_image I will use a 50hz tone, then.
