my sister has a '00 Xterra, and I have made a custom sub box for the rear of it.  she doesn't want to to put up the money for a new head unit, so I have no RCA cables for the sub amp.

the Xterra has rear panel speakers, so I could use the rear speaker wire to connect an amp with high-level inputs.  I want to keep the rear speakers connected, but is that going to be possible?  can i just split the rear speaker wires into two wires each, keeping the speakers wired, but peeling off to feed the source to the amp?

Yes, taht is what you will need to do. Just piggyback the two rear speakers into the amp's high level inputs.
