I want to build a custom box for my Alpine R 12" Sub, that will fit in my Highlander with least space intrusion in the trunk and most aesthetic/factory look. What kind of materials should I use to get best sound quality? I don't care about spl. I have too much of it.
Plastic? Wood? Plexiglass? Also does the shape of the box matter? I know that the volume matters, such that I should try to stay within the required cft requirements. So if I have the right volume (cft) of air in the box, and the shape is not a cube, will it sound good?

Any info would be appreciated, preferably from knowledgeable folks. Thanks.

use a combinatino of mdf and fiberglass to create an enclosure that suits you. the size doesn't matter and fiberglass and mdf are both great materials to build enclosures with. just make sure you get the internal volume correct.
