Yep, your on the right path. One thing you might wanna try instead of a round port is maybe going with a slot port design instead. It's alil more easier to make and you don't have to worry 'bout the chance of port noise you get from using round ports.

tuning to 50hz is a little high i would think....maybe try between 30hz and 40 hz but thats just my oppinion you may have reasons for 50hz. you should be really happy with those subs

how would i go about making a sloted port??? How could i tun it to somewhere in the range of 42-50 hz

You are going to be blown away with that box.  The loudest system iv heard so far is from 2 type-r 12's ported to 50hz.  but i would reccomend going down to about 40 hz its a more rounded sound.

If you've already calculated that you need 3" round ports at 5.5" long, buy pre-made flared plastic ports and mount them.  No need to change your design.

Polyfill will increase the effective enclosure volume.  about 1 lb per 1 cuft will incerase the effective volume by 10% (0.1 cuft).  Yes, you must take this into consideration.
