OK, I hooked up the Farenhiet FMT-40D wireless FM transmitter to a gorund and positive and everything was working fine, I turned the car off and turned it back on and the unit did not power up again. I checked the fuse which was fine, but did replace it to test, but no luck. I opened the unit up and made sure that power was reaching the board inside, and tested it with a voltmeter and the power was reaching the circuit board were the + and - were soldered to, but it would still not power up. Can someone please help me?

I don't know anything about that unit, but in general if you have something connected correctly and it stops working, it may be defective.  Take it back for warranty replacement.
