I have a 2001 Cadillac Deville, and I am thinking of getting rid of the freeair woofer thats in the car, and putting in a 10" aftermarket sub, I would like to mount a enclosure to the rear deck, could i do this without too much vibration?

If you have the space you can design an sealed enclosure that will mount to the rear deck and have the subs firing up towards the back window. If you seal the enclosue correct and also the area around it you will eliminate almost all the sound going into your trunk. This will of course stop the rattling in your trunk and increase the sq of your system.  I don't know if you would be able to pull this off just but its just an idea.
