I am building an enclosure & I was wondering: someone told me that painting the inside of the enclosure with some kind of latex based paint will help the sound damping. Is this true. Also the enclosure is going to have a slot port. Would it be better if there are 2 ports or just 1 port tuned to the same frequency or does this really matter?

heres an answer to part of your question taken from one old old old thread;
"go wo wal-mart and get a can of bed liner spray. This is a cheeper for of the line-x, Rhino liner stuff and works great. I use this stuff constantly inside every box that I build to both seal the box and stop resonations. Plus it is not that messy or greasy like the undercoating is. You can also spray it up to about a 1/4 inch thick and it wont come off."
as for the ports: It depends on the subs and the box dimensions. If the slot is tuned to both subs then there should not be a problem. Take time to round the edges of the slot, both outside and inside the box, to let the air moving through the box move smoothly. Best suggestion, "think airodynamicly".
