enclosure question

i am curious if anyone here is familiar with the lexus gs300/400/430 models. i am going to be running four 10w6v2 sub either fiberglassed in te spare tire well or towards the front of the trunk right behind the rear seat. my questions to you all is,

if i build a box behind the seat in the front of the trunk will i make better gains if i point the speakers away from the passanger area towards the trunk lid of down towards the floor.

my second question is, since i know the trunk is like a big steal box, what is the best way to get the sound into the passanger area. i already intend on removing the stock sub so that will open up a 10 inch hole between the trunk and the passanger area. will this hole be enough or should i open up some more vents.
