How do I use a digital multi meter to test a speaker wiring circuit? What settings and readings should I be testing for?

Are you trying to check if your speakers are good?  Put the meter on resistance (ohms), and touch the two wires behind the radio for the speaker you want to test.  Most (not all) car speakers are generally around 4 ohms. Now just because they read 4 ohms doesn't necessarily mean they're good- they can still crackle, etc.
My ex once told me I have a perfect face for radio.

You can set it to ohms and measure the resistance, my meter allows me to send a signal out and lets me vary the hz. But ya need to be more specific
