What Frequency to use when setting gains?

Does it matter what frequency I use to set the gain of my subwoofer amp. I know it has to be below 80 Hz so the LP filter doesn't kick in, but does it matter if I use a 50hz tone, 70 hz, 30 hz, or what frequency of a tone. Does it matter.

50 Hz will be about as robust as they get.
Build the box so that it performs well in the worst case scenario and, in return, it will reward you at all times.

I feel like an idiot. My mind blanked I had to look robust up What Frequency to use when setting gains? -- posted image. I will use a 50hz tone, then.
